چکیده
Respirable cotton dust, implicated in the pathogenesis of byssinosis, contains a number of bioactive compounds. These include lipopolysaccharide (LPS), tannins, bacterial peptides, byssinosin, iacinilene C, and 1,3-beta-D-glucan. The exact aetiological agent of byssinosis in such dust has not been definitively identified nor has its mechanism of action on lower lung surfaces been determined. In the present study 1,3-beta-D-glucan, Enterobacter agglomerans LPS, and ovine pulmonary surfactant were mixed in varying combinations. After incubation, their characteristics were determined by sucrose density centrifugation, TLC, and carbohydrate analysis. Precipitates were found in mixtures containing surfactant-glucan and surfactant-glucan-LPS, but not in surfactant-LPS. Precipitates were not seen in the surfactant, LPS, and glucan controls. The formation of a precipitate did not increase the density of the surfactant glucan mixture when compared by density gradient centrifugation with the surfactant control. The interaction between surfactant and glucan was analysed by molecular modelling. The energy of a surfactant-glucan complex (60.07 kcal/mol) was calculated to be much lower than the sum of glucan (47.09 kcal/mol) and surfactant (30.98 kcal/mol) when added separately. The results indicate that 1,3-beta-D-glucan does interact with surfactant and this complex may play a part in the pathogenesis of byssinosis by altering lung physiology maintained by pulmonary surfactant.

Surfactant Protein A in Cystic Fibrosis: Supratrimeric Structure and Pulmonary Outcome
BACKGROUND The state of oligomerization of surfactant associated protein-A (SP-A) monomers differs between individuals. This likely affects SP-A's functional properties and could thereby influence clinical status in patients with lung diseases. In this study we focus on SP-A structure in cystic fibrosis (CF) compared to both healthy subjects and disease controls. METHODS SP-A composition and ...
متن کاملThe effect of Environmental exposure to some chemical solvents on DPPC as important component of lung surfactant: an ab initio study
One of the main components of lung alveoli is surfactant. DPPC (Dipalmitolphosphatidylcholine) is thepredominant lipid component in lung surfactant that is responsible for lowering surface tension in alveoli in thisarticle. We used a very approximate model with computational method of Ab initio to describe the interactionsbetween DPPC as important component of lung surfactant and some chemical ...
متن کاملBinding of pulmonary surfactant proteins A and D to Aspergillus fumigatus conidia enhances phagocytosis and killing by human neutrophils and alveolar macrophages.
To determine whether the lung surfactant proteins A (SP-A) and D (SP-D) are involved in the initial protective immunity against opportunistic pulmonary fungal infections caused by Aspergillus fumigatus, we performed a series of in vitro functional studies to see if SP-A and SP-D enhanced binding, phagocytosis, activation, and killing of A. fumigatus conidia by human alveolar macrophages and cir...
متن کاملInvestigation of histopathological and radiological effects of surfactant treatment in an experimental female rat model of lung contusion
Objective(s): Pulmonary contusion (PC) is a clinical entity that often accompanies blunt traumas. We aimed to investigate the radiological and histopathological effects of surfactant treatment in an experimental rat model in which lung contusion was formed by blunt thoracic trauma.Materials and Methods: 50 female Sprague-Dawley rats were...
متن کاملAntibody-mediated inhibition of dextran-sucrose-induced agglutination of Streptococcus mutans.
Immune sera to strains of Streptococcus mutans were found to inhibit agglutination of bacterial suspensions to which either high-molecular-weight dextran or sucrose was added. This inhibitory activity was shown to be mediated by antibody of the immunoglobulin G class. A semiquantitative assay was developed which demonstrated cross-inhibition of dextran/sucrose-induced agglutination among severa...
